No findings were extractable from the material analysed.
Safety was not reported in the material analysed. Check the source before drawing any conclusion about harm.
The source did not state who this applies to in practice.
Graded across the dimensions that decide whether you should act, each from what the source actually supports. There is no single score, and where a dimension was not assessed it says so.
This record has not been graded across any dimension yet. Treat the label above as provisional and read the source.
What is missing. This record has no bottom line, key findings, reported figures, evidence dimensions. That is a gap in the analysis, not a judgement about the study.
CASTOR1, a cytosolic arginine sensor and regulator of mTORC1 signaling, is identified here as a tumor suppressor in KRAS -driven non–small cell lung cancer (NSCLC). In a KRAS G12D mouse model, CASTOR1 deletion enhances tumor initiation and progression, accompanied by increased proliferation. Although CASTOR1 is rarely mutated or deleted in human cancers, its expression progressively declines during tumor progression and is associated with increased AKT activation and CASTOR1 phosphorylation, consistent with functional inactivation. Loss of CASTOR1 results in constitutive mTORC1 hyperactivation, impaired arginine-dependent regulation of mTORC1 signaling, elevated p-4EBP1 and p-S6 levels, increased AKT and ERK activation, genomic instability, and resistance to KRAS G12D inhibitors. Tumors and tumor-derived organoids deficient in CASTOR1 are resistant to KRAS inhibition, but this resistance can be overcome by cotargeting mTORC1 or dual PI3K/mTOR inhibition. These findings identify CASTOR1 as a critical link between nutrient sensing and oncogenic signaling and reveal its functional inactivation as an important mechanism contributing to KRAS -driven NSCLC progression and therapeutic resistance. The results further support rational combination therapies to overcome KRAS inhibitor resistance in CASTOR1-low NSCLC.
